Attempt has been made to identify Donaggio positive substances in urine after exercises (severe running). Results obtained were as follows; 1) The substances having the high titer of Donaggio reaction, were probably derived from glycoproteins, which include α_1-globulin, and small amounts of α_2-globulin, by cellulose-acetate electrophoresis. 2) This α_1-globulin nearly composed of α_1-acid-glycoprotein (so called α_1 AG) and other α_1-glycoprotein were not found, namely tryptophan poor α_1-glycoprotein(α_1 T), α_1-anti-trypsin (α_1 AT), and α_1-anti-chymotrypsin (α_1X) by immuno double diffusion. One of the α_2-globulin on Donaggio positive substances was found to be α_2HS-globulin (α_2HS). Single radial immunodiffusion (SRID) and electro-immunodiffusion (EID) were used to measure the levels of Donaggio positive substances (α_1AG and, α_2HS) in urine before and after exercise, levels of α_1AG, α_2HS in urine measured by SRID were almost similar to those by EID. 3) Both α_1AG and α_2HS in exercised urine were slightly decreased heating (100℃ 10 minutes) after, being acidified with acetic acid (pH 5 or pH 4). The lower pH indicated, the lower α_1AG and α_2HS level.) The treatments were (only boiled and filtrated) described by Donaggio in his original manuscript. Data indicates that adjusting urine to pH 5 is recommended.
