In bone resorbing osteoclasts proton-pump ATPase is known to mediate the acidification of the extracellular ruffled border and intracellular organelles. We have tried to visualize the acidic compartment at various states in cultured osteoclasts by an indirect immtunocytochemistry for DAMP. The resorbing osteoclasts showed the polarityindicating the ruffled border and resorptive pit which were an acidic compartments. The eroded bone surface still maintain the acidic pH after removing osteoclasts. Both baifilomycin and acetoazolamide decreased the acidity of the osteoclasts. In conclusion, DAMP immunocytochemistry demonstrated that not only the intracellular organelles but also the extracellular resorptive lacuna were acidic.
