Accumlation of Nucleic Acids in Isolated Pupal Abdomens of the Silkworm, Bombyx mori (Lepidoptera : Bombycidae) Infected with Nuclear Polyhedrosis Virus

Nucleic acids from NPV infected and uninfected isolated pupal abdomens of the silkworm, Bombyx mori, were analyzed using MAK column chromatography. The elution profiles of nucleic acids from uninfected isolated abdomens were similar to those from intact pupae during histolysis, and showed 6 peaks which corresponded to tRNA, 5 S RNA, X_1-DNA, X_2-DNA, normal DNA and rRNA. These nucleic acid components in isolated abdomens decreased with age, indicating that considerable degradation of nucleic acids was taking place in these abdomens in an arrested state of development. Although the number of peaks was the same regardless of whether or not isolated abdomens were infected with an NPV, NPV infection caused marked acuumulation of RNA and DNA components. The increased accumulation of cellular RNA indicates that NPV infection is accompanied by stimulation of cellular RNA synthesis and hence implies that some product of NPV genes acts on cellular genes to stimulate this synthesis.
