We examined the mechanism of apoptosis induced by staphylococcal enterotoxin B (SEB) in cultured monocytes. Apoptosis of monocytes was enhanced by SEB. Enhanced production of soluble Fas ligand (sFasL) was induced by SEB, but not by IFN-γ produced by SEB stimulation. SEB treatment induced the activation of caspase-3 and -8, and pretreatment with z-VAD-FMK, a broad-spectrum inhibitor of caspases, prevented the induction of apoptosis. Pyrrolidine dithiocarbamate (PDTC), a potent inhibitor of NF-κB, slightly enhanced and markedly enhanced apoptosis induced by SEB and IFN-γ, respectively. In monocytes, NF-κB was constitutively activated, and SEB further activated NF-κB, which was inhibited by pretreatment with PDTC. These observations suggestthat SEB has dual effects on apoptosis of monocytes : anti-apoptotic action through NF-κB activation and pro-apoptotic action through sFasL release from activated T cells.SEBapoptosiscaspaseNF-κBmonocyte
