We investigated factors that enhance chemiluminescence (CL) detection of active oxygen generated by imidazole catalysis from dopamine using a flow injection system and peroxyoxalate chemiluminescence. The factors investigated were the sources of imidazole, the effect of perchlorate, and the types of chemiluminescence detection systems (luminol/peroxidase, peroxyoxalate chemiluminescence (TDPO/perylene), luminol/ferricyanide, lucigenin, pyrogallol, purpurogallin, luminol). Rank order of light intensity for the imidazole sources was Katayama (100%)>Kishida (72)>Wako (52.7)>Kanto (50.8)>Nakarai (43.5). Perchlorate (0,10,25,50 and 100mmol/L) incubated with the imidazole solution containing dopamine decreased the background light, and perchlorate (50 and 100mmol/L) elevated the signal to background ratio 16.4% and 26.6%, respectively. Rank order for the CL detection systems was luminol/peroxidase (31,700 RLU)>peroxyoxalate chemiluminescence (854)>luminol/ferricyanide (530)>lucigenin (128)>pyrogallol (6.6)>purpurogallin (3.1)>luminol (2.6). The imidazole from Katayama, the addition of perchlorate, and the detection by luminol/horseradish peroxidase chemiluminescence were effective for enhanced CL detection of active oxygen.
