In case of human mitochondrial DNA (mtDNA), the deletion of mtDNA with 4977-bp length is called common deletion, because it has been frequently observed. We examined the time course of the appearance and disappearance of deleted mtDNA caused by endurance exercise. Blood was sampled from in two individuals (41 year-old male; and a 21 year-old female) before and after three sets of exercise, and mtDNA was analyzed using the nested PCR procedure. Before exercise, a common deletion of mtDNA was not identified in either individual. A common deletion of mtDNA appeared at two and five days in one subject, but disappeared seven after completing the exercise. A common deletion appeared in the blood mtDNA three days after exercise, and again two days later in the other subject. These data indicate that a common deletion of mtDNA appeared two to three days after endurance exercise. Furthermore, this deletion was dynamic, since it disappeared after a period of rest.
