Modification of Blood Flow After Injection of Hydralazine in Tumors and Normal Tissues.

An investigation was carried out the conditions which increase blood flow in normal tissues surrounding a tumor, by selectively reducting blood flow in the tumor tissues with Hydralazine (HYD), a vasodilator. HYD was administered to 141 tumor bearing rats at dosage levels of 2.5,5.O, and 10 mg/kg. 30 and 60 minutes after treatment with HYD, 125-I labeled microspheres were administered. Intra-tissue blood flow was estimated from radioactivity in the tissues. Following administration of HYD, blood flow in the normal tissues increased in muscles with a rise in cardiac output, but decreased in the other tissues. In all of the tumors transplanted to rgans such as the liver, blood flow declined sharply. In tumor transplanted to the muscle, blood flow showed an inverse proportional decrease to increase in tumor size in the HYD-untreated group. In the HYD treated group, blood flow decreased from a mean pre-treatment level of 0.15 ± 0.02 ml/min/g to a fixed mean level of 0.05 ± 0.01 ml/min/g, regardless of the tumor size. These findings showed that treatment with HYD could reduce the intra-tumor blood flow without relation to the tumor size. It is expected that HYD treatment will be applicable to hyperthermia.
