T cells require costimulatory signals through CD28 by binding with B7-1 (CD80) to induce full activation. Previous studies in mice have shown that inoculation of B7-1 positive tumor cells led to the tumor regression, and that immunization with B7-1 positive tumor cells induced protection from further challenge with the tumor even without B7-1 expression. In the present study, role of B7-1 was investigated in human allogeneic system in vitro. A squamous cell carcinoma line, OSC40, which is MHC class I positive, and MHC class II negative, was made immunogenic after transfection with B7-1 gene. Allogenic peripheral blood lymphocyte (PBL) pre-activated with phytohemagglutinin proved to respond efficiently to OSC40 cells transfected with human B7-1 gene, when estimated by TNFα production and proliferation assays. The results suggested that B7-1 expression augmented immunogenecity of tumor cells with low immunogenecity. Furthermore the findings suggested that introduction of B7-1 gene in tumor cells may be useful to generate tumor specific cytotoxic T lymphocytes (CTLs). The biological significance of B7-1 expression in tumor immunology was discussed.
