The light-induced oxidation of plastocyanin and its dark reduction in 180 S were measured at 580 nm. An initial rapid and followed slow oxidation of plastocyanin upon illumination of 180 S were observed. The initial rapid oxidation of plastocyanin in 180 S was enhanced by addition of 110 P. The slow oxidation of plastocyanin in 180 S was diminished and slow reduction of plastocyanin was observed during illumination by addition of chloroplasts or 20 P. The light-induced slow reduction of plastocyanin in the presence of 180 S and chloroplasts was abolished by addition of o-phenanthroline, while the light-induced rapid oxidation was not affected. After the light was turned off, a rapid and followed slow reduction of plastocyanin in 180 S were occured and reached the initial oxidation-reduction level before illumination. Such recovery of the light-induced oxidation of plastocyanin in 180 S during dark may be related to the presence of a low molecular substance (s) such as CRS or FRS.
