The protein associated with fluorescent substances was extracted from spinach leaves with 0.05M Tris-HCI, pH 7.8. The protein was purified by ion exchange chromatography on DEAE cellulose, gel filtration on Sephadex G-50 and fractionation of ammonium sulfate. The contaminating flavin compounds were removed by Sephadex G-50 column. The absorption and the fluorescece spectra of the protein were similar to those reported for the pteridine compounds. The enzymatic treatment with papain (Nagase) removed the fluorescent substances from the protein. The Rf values of paper chromatogrphy of the fluorescent substances did not identical that of the authentic pteridine compounds. It stimulated photophosphorylation of isolated chloroplasts by the addition of the fluorescent substances in the absence or presence of electron acceptor. But the photophosphorylation of isolated chloroplasts catalized with PMS was not affected by the addition of the fluorescent substances. These results might suggest that the fluorescent substances act as electron acceptor between photosystem I and photosystem II.
