Several culture conditions were investigated, aiming to get higher Perilla pigment production with Perilla callus tissues (2-6 L and 2-3-2 D) having high pigment producing capacity. It was revealed that blue light was most effective of three light sources (blue, red and white) for the pigment production, and that the promotive effect of blue light increased as the intensity of the illumination decreased. Both two strains also produced a considerable amount of Perilla pigment even in darkness. The optimal regions of α-NAA and kinetin in a culture medium were 0.1-0.2ppm and 1.5-2.5ppm, respectively. Four weeks' static culture and 2-3 weeks' suspension culture produced the highest yields for the pigment production, respectively. Under the best culture condition mentioned above, the pigment production in Perilla cell and tissue culture has been elevated to ca. 5.5mg pigment/g fresh weight. This value seems to be equal to that of the intact plant leaf. Red pigments extracted from the callus tissues were found by paper chromatographic analysis to be similar with those (anthocyanin pigments) from the intact plant leaf, and the aglycone moiety of the anthocyanin pigments was identical with cyanidin.
