Callus induction from segments of Perilla plants, growth and pigment formation of the callus tissues were examined under various culture conditions. Callus tissues were easily induced from segments of sterile Perilla seedlings in the presence of auxin (α-NAA or 2,4-D) and kinetin (0.2∿2.0ppm). The growth of Perilla callus tissues (RT-4 and BT-6) was strikingly promoted when they were cultured at a limited concentration (1.0∿2.0ppm) of α-NAA and 0.2∿2.0ppm cytokinin (kinetin or benzyladenine) in culture medium. The optimal concentration of sucrose was 3%. Blue light illumination seemed to stimulate the growth of red Perilla callus tissues (RT-4). Red purple pigment formation was recognized in the callus tissues induced from leaf segments of red Perilla plants under blue light illumination. High pigment producing strain was obtained from the above callus tissues by selection.
