To elucidate the pathogenesis of vascular changes caused by transarterial infusion of anticancer drugs, the rat hepatic artery and mesenteric artery were examined morphologically and biochemically after cisplatin-lipiodol infusion. Severe degenerative changes were found in endothelial cells as well as medial smooth muscle cells in hepatic and mesenteric arteries. Such changes were milder in the artery in the group administered cisplatin-lipiodol and vitamin E (vitamin E group) than in the group administered cisplatin-lipiodol (cisplatin group). Superoxide dismutase, a superoxide anion scavenger, was higher in the vitamin E group than in the cisplatin group. Furthermore, Na/K-ATPase, a membrane-related enzyme, was also higher in the vitamin E group. These findings indicate that the cisplatin-induced arterial changes were caused by lipid-peroxidation due to free radicals and thus prevented by vitamin E administration.
