The target antigen, 54 kD glycoprotein (gp54), involved in the anti-tubular basement membrane (anti-TBM) antibody-mediated tubulo-interstitial nephritis, and localized dominantly to the basement membrane of proximal tubules was characterized. gp54 was isolated from collagenase-digested (CD) bovine TBM by column chromatagraphy and further elution from blotted membranes. Amino acid analysis showed that the gp54 was non-collagenous in nature. The NH_2-terminal amino acid sequencing of gp54 revealed that the gp54 was a unique basement membrane component. The nephritogenicity of gp54 was demonstrated by immunizing guinea pigs (strain XIII) with purified gp54,and producing anti-gp54 antibody and tubulo-interstitial nephritis. Anti-gp54 antibody, affinity-purified from sera of patients with anti-TBM nephritis, anti-gp54 peptide antibody, and antibody eluted from guinea pigs immunized with gp54 bound to 54 kD and 48 kD components of human CD-TBM. mRNA was prepared from rabbit kidneys and concentrated by the sucrose density gradient centrifugation method. ^<35>S-Me-thionine-labeled proteins translated from the rabbit renal mRNA were incubated and immunoprecipitated with anti-gp54 antibody, revealing mainly 54 kD and 48 kD components. A competition test using purified gp54 showed that anti-gp54 antibody was cross-reactive with 48kD component. Dot blot hybridization with an oligonucleotide probe for gp54 revealed a positive signal in cDNA libraries mada from adult and fetal kidneys. These findings indicate that 54kD and 48kD have similar antigenecity, but are encoded by different mRNA.
