Eosinophils are considered to be related to the pathogenesis of bronchial asthma in the inflammatory reaction. To extend our understanding of eosinophils in relation to bronchial hyperresponsiveness as well as bronchial mucosal damage, we examined the effect of inhaling human eosinophils on bronchial hyperresponsiveness and bronchial damage on ovalbumin (OA)-sensitized guinea pigs. Prior to OA challenge, eosinophils, eosinophils+platelet factor 4 (PF4), PF4,or sonicated eosinophils, were inhaled by OA-sensitized guinea pigs. Whereas the guinea pigs treated with inhalation of medium alone only showed immediate asthmatic response (IAR) after OA challenge, the guinea pigs treated with inhalation of eosinophils, eosinophils+PF4,PF4 or sonicated eosinophils showed not only IAR but also late asthmatic response (LAR). Frequency of bronchial ciliary beats (CBF) was diminished by eosinophils, and more greatly diminished by eosinophils+PF4,PF4 or sonicated eosinophils. In conclusion, greater bronchial mucosal damage was caused by eosinophils attracted and activated by eosinophil chemoattractants such as PF4 than by eosinophil alone, resulting in bronchial hyperresponsiveness. This suggested that bronchial hyperresponsiveness might be closely related to bronchial mucosal damage through allergic inflammation mainly due to eosinophils.
