To clarify the interaction of splenocytes and grafts when grafts are trapped in the spleen after autologous bone marrow transplantation in cancer patients, experiments using mouse cells were conducted in vitro. Female DBA2/Cr mice aged 8 to 9 weeks and tumor cell lines L1210,P388D_1 and K562 were used. After L1210 cells and mouse splenocytes were cultured, mouse non-adherent bone marrow cells were added and culturing was continued. The resulting supernatant was separated from the culture broth (supernatant I). Using supernatant I, non-adherent bone marrow cells were cultured, and the resulting supernatant was collected (supernatant II). Factorial assay revealed that supernatant I contained interleukin-3 and interleukin-6. Each tumor cell line was cultured using supernatant II. Supernatant II was found to contain a species-specific tumor growth suppressor that significantly (P<0.001) suppressed the growth of L1210 cells and P388D_1 cells in comparison with controls, but did not suppress the growth or K562 cells. It is inferred that a similar tumor growth suppressor is also produced when grafts are trapped in the spleen in vivo.
