The analog of muramyl dipeptide, MDP-Lys (L18) is resistant to bacteria and acts as an immunoadjuvant. The effect of MDP-Lys(L18) on immunoability was examined, using NZB/WF_1 mice, a model for human autoimmune disease. MDP-Lys(L18) induced the production of IL-1 in spleen cells of NZB/WF_1 and BALB/c mice used as controls, and had an adjuvant effect in vitro. However, it had no effect on IL-2 production in vitro. MDP-Lys(L18) increased the number of macrophages in vivo. Though IL-1 and IL-2 were produced in NZB/WF_1 mice at a lower rate than in BALB/c mice, the lymphokine production increased in treated groups in both types of mice, and that in treated young NZB/WF_1 mice reached nearly the level as in BALB/c mice. The IL-2 response was lower in NZB/WF_1 mice and was significantly higher in treated groups than in non-treated groups. These findings suggest that MDP-Lys(L18) improves the immunological response of NZB/WF_1 mice when T cells and macrophages are stimulated for immuno-modulation.
