Corneal fibroblasts are surrounded by regularly oriented collagen fibers. To investigate the role of corneal fibroblasts during corneal wound healing, the changes in the phagocytic activity of corneal fibroblasts for latex beads were studied in vitro using three-dimensional collagen-gel matrix. Phagocytic activity of corneal fibroblasts cultured both on a plastic culture dish and in a collagen-gel increased linearly with incubation periods up to 12 hours. After each incubation period examined, corneal fibroblasts cultured on plastic dishes showed higher phagocytic activity than those cultured in a collagen-gel. Phagocytic activities of corneal fibroblasts decreased with the concentration of collagen. Incorporation of ^3H-thymidine of corneal fibroblasts cultured in a collagen-gel also decreased with the concentration of collagen-gel. Phagocytosing corneal fibroblasts incorporated more ^3H-thymidine than non-phagocytosing cells. These results demonstrated that cultured corneal fibroblasts have phagocytic activity even in a collagen-gel and that the phagocytic activity of corneal fibroblasts is modulated by the presence of extracellular collagen.
