In order to investigate morphologically very early stage damage and the recovery of myocardial fibers in relation to electrolyte and water contents in myocardium, experimental myocardial infarcts were produced by the administration of isoproterenol (200 mg/kg) to Wistar rats weighing 260±10g. Electrolyte and water contents were examined by an atomic absorption spectrophotometer. Histologic examinations were performed on preparations of HE stain, antimyoglobin, and anti-actin antibody reactions. The later two reactions were estimated by a color image analyzer. At 30 minutes after the injection the Ca and water contents of the myocardium were increased, and Mg was decreased. Also, the anti-myoglobin and anti-actin antibody reactions of myocardial fibers disappeared in subendocardial regions. These facts indicate myocardial damage during this period. The findings of damages reached a maximum level at 12 hours after the injection. The normal positive areas of myoglobin and actin reaction occupied 26.9% and 25.1%, respectively, in the cut surface section of the heart. The sections stained with HE showed consistently intermixture of eosinophilic degenerative and coagulative necrotic fibers. At 3 days after the injection the eosinophilic fibers were not revealed, but well demarcated coagulated necrotic fibers remained, and a positive reaction with anti-myoglobin and anti-actin antibody had diffusely recovered to normal intensity, except for the necrotic fibers. The normal positive area increased to 83.3% for anti-myoglobin and 76.6% for anti-actin reaction. The results indicated that (1) chronological changes of Ca, Mg, and water contents correlated well with the morphological changes of myocardial fibers, and (2) 56.4 and 51.5% of the damaged fibers had recovered as estimated by anti-myoglobin and anti-actin antibody reaction, respectively. These observations on this experimental model may be useful in examining the recovery of damaged myocardial tissue and in estimating the effects of therapeutics.
