The author studied the effects of methoxyflurane having potent analgesic property and halothane having less one on the binding of various opiate ligands to rat brain membrane. Furthermore, effects of these two anesthetics on methionine-enkephalin (Met-EK) content in the brain and spinal cord were studied. The binding affinity of[^3H]-Met-EK(δ) was reduced by both anesthetics in the same manner, but the receptor density was not significantly affected. Methoxyflurane decreased the binding affinity but increased the receptor density of ^3H-dihydromorphine(μ), while halothane had little effect on both affinity and density. Methoxyflurane also decreased binding affinities of[^3H]-ethylketocyclazocine (κ) and ^3H-SKF-10047(σ), but halothane did not decrease the affinities significantly. These effects of the anesthetics completely disappeared when the anesthetics were blown away from the test tube by unsealing and ventilation. On the other hand, these two anesthetics did not alter Met-EK content in the brain and spinal cord, significantly. Thus the present study suggests that analgesic property of the anesthetics may be relevant to the opiate receptors and that the increase in number of receptor may be important for the analgesia of anesthetics.
