Replication of Mitotic Chromosomes and a Karyotype Analysis of Vicia faba by RHQ Staining Method after Incorporation of BrdU

The replication patterns of mitotic M chromosomes and a karyotype analysis at mitotic metaphase of Vicia faba are reported by using RHQ staining method with BrdU incorporation following the partial synchronization of mitotic division by the HU treatment. RHQ staining method is modified for Vicia faba as follows. BrdU concentration is 100μg/ml and staining time of Giemsa is 4 min. The optimum condition by HU treatment is decided and used as 1.25mM for 20.0 hours and recovery time is 12.0 hours. Almost all M chromosome pairs showed same replication patterns and they were divided into the three patterns that were replicated at early, middle and late periods of S phase. Though all S chromosomes resemble each other morphologically, each chromosome pair was distinguished easily by appearance of RHQ bands.
