In a GvH reaction system which utilized the relation of genetic constitution, production by injecting BDF_1 hybrid animals with immunological competent cells from one of the parent strains (C57BL/6 or DBA/2 strain) against which the BDF_1 as recipient is immunologically defenceless, we showed that several adenine nucleotides and N^6-purine analogues had the stimulative effect. In normal BDF_1 mice not injected with parental spleen cells, all compounds have not been shown to be significantly different from those in control mice (PBS-injected mice). It was clearly found that in chemical form levels of purine compounds, several adenine nucleotides have 40 to 90% increase on spleen index in GvH mice, but that adenosine, guanosine and several guanine nucleotides do not. GvH stimulative effect-possessing adenine nucleotides included 3' : 5' c-AMP, dBut c-AMP and 8-Br c-AMP but not 3' : 5' c-GMP. It has not been known that noncyclic nucleotides had the effect on immunological responses in vivo. Also we found that N^6-purine ribo-nucleosides except for N^6-furfryladenosine exhibited the stimulative effect on GvH reaction.
