A new detection method for ligand–protein interactions was constructed using interaction-dependent native chemical ligation and enzyme reconstitution (IDNCL-ER) by employing ribonuclease S. Using a coiled-coil peptide pair as a model of ligand–protein interactions, we determined that IDNCL-ER can work well to detect the peptide–peptide interactions by enzymatic activity.
