Recently, it is noticed that the patients of psittacosis have been increased in number and some deceased cases have been found. Therefore, the check system for this disease is pressingly needed. The antigens for the serological surveys must be prepared personally in our own laboratory, because they have not been on the market. This report concerns with two studies on Chlamydia psittacosis, the one is a preparation of antigens employing for hemagglutination inhibition (HI) and complement fixation (CF) test, and the other is the serological survey results of the subjects in Aichi prefecture. 1. Preparation of the antigens HA antigen for HI test was: prepared as follows; Seven to eight day old embryonated eggs were inoculated into the allantoic cavities with 0.2ml/egg of A-8 strain, diluted with saline in 10-2 to 10-4 (originated from human patient, ELD50 10-8.1) and incubated at 35°C for 5 to 8 days. For the CF test, allantoic fluids of 7 to 8 day old eggs which were inoculated with 0.2 ml of the same strain (in dilution of 10-2) into the allantoic cavities and were incubated at 35°C for 6 days, were prepared, and those fluids, revealed the value of 1: 16 to 1: 64 in HA titer, were used as CF antigen. HA antigen was well preserved for 6 months at-80°C and CF antigen for 3 months at 4°C. 2. Serological surveys Both of HA and CF tests against psittacosis were surveyed on 180 subjects of the age group of 1 to 40 years old (including the healthy subjects and the ill patients without respiratory infection) in Aichi prefecture. CF positive subjects were observed in 3 of 180 (1.7%) at the titer of≥1: 8 and one of 180 (0.6%) was HI positive at the titer of≥1: 16. Forty-five paired sera of the patients with respiratory diseases or febrile diseases were also examined, one subject among febrile patients showed a significant increase of CF antibody.
