A Signal Extraction Method for Cortical Intrinsic Optical Changes Driven by Visual Stimulation

Optical imaging based on intrinsic signals is widely used as one of the methods for visualizing the cortical cell activity induced by external stimulation. However, the evoked optical change is usually much smaller compared to the background optical intensity. Separation of the stimulus dependent optical change from the background is critical for the application of this technique. As the background optical change is generally independent of the stimulus presentation, in this study, we propose a signal extraction method based on the temporal relation between stimulus presentation and recorded optical changes. We have applied this method to visualize the orientation column in cat primary visual cortex. The results demonstrate a strong ability to separate stimulus-driven optical changes from background noise such as that caused by the movement of blood vessels or the cortical surface.
