Effect of Metyrapone (SU 4885) on renin-angiotensin-aldosterone system of fasting rat

We studied the effect of Metyrapone on the excretion of 17-OHCS, plasma renin activity (method of Skinner), and plasma aldosterone level in the fasting rat. The Wister-strained rats were injected Metyrapone (SU 4885, Ciba) 10 mg/100 g daily from the beginning of rat fasting for three days. The excretions of 17-OHCS decreased in the fasting rats, and also the daily additional injections of Metyrapone into the fasting rats similiarly did not raise the excretions of 17-OHCS. The plasma aldosterone levels were increasing as the course of time of fasting day, and also the daily additional injections of Metyrapone into the fasting rats did not decrease the plasma aldosterone levels. It was concluded that fasting might produce the hyporesponsiveness of adrenocortical 11-hydroxylation to Metyrapone, because the injection of Metyrapone into fasting rat could not affect the values of 17-OHCS and plasma aldosterone.
