Tumor Promoting Potential of Suspended Particulate Matter Collected in 13 Size Classis

To study the tumor promoting potential of suspended particulate matter (SPM), inductions of transformed foci by 13 size-fractionated airborne samples, which were collected by an Andersen low pressure impactor, the transformation activities were measured using Bhas 42 cells (Bhas promotion assay) . The numbers of transformed foci per unit mass for the samples of less than 2.5 μm in diameter were generally larger than those for the samples of more than 2.5μm in diameter. The highest promoting potential per unit mass, about 7 times the median value, was observed in the sample of less than 0.06μm in diameter. Though it represented the smallest portion of the total particulate mass, this high promoting potency gave it the fourth highest relative promoting potential. The promoting potential per unit air volume in three samples of around 0.76μm in diameter was higher than those in the other samples, and which approximately matched their predominance on a weight basis within the whole particulate matter. These results also showed that most of the total promoting potential (ca.70 %) was in the fine fractions (<1μm diam.) . Concerning mutagenicity, size-fractionated particulate air samples collected in Tokyo one year prior to the present samples were assayed in an Ames test with<I>S. typhimurium</I>YG 1024. These data showed that the smaller sized particles are the most active mutagens. The present experiments on size-fractionated particulate matter demonstrate that smaller sized particles within air samples have the highest<I>in vitro</I>mutagenic and promoter activity in the assays used. Therefore it should be necessary to pay close attention to the association of air pollution with cancers other than respiratory cancers because the tumor promoters and initiators in ultra fine particles may penetrate the body directly through the blood vessels via the lungs.
