Thirty-two patients with squamous cell carcinoma of the head and neck, 17 of whom had received no prior therapy, were given a combination chemotherapy (VPCP regimen) consisting of vincristin (V), 1mg/day on day 1; peplomycin (P), 10mg/day on day 2; cisdiamminedichloroplatinum (II)(C), 80mg/day with mannitol diuresis on day, 3 ; and peplomycin (P), 5 mg/day on days 4-8. In principle, the regimen was mepeated at 3 week intervals, for a maximum of three cycles. Among 29 evaluable patients, the overall response rate was 59% with 6 (21%) complete responders and 11 (38%) partial responders. Among 17 patients without prior therapy, the response rate was 65%. Toxic effects were within acceptable limits and there were no fatalities. A majority of the patients experienced anorexia, nausea, and vomiting, but those symptoms were self-limited. Bone marrow suppression occurred in only four patients, and renal dysfunction was not seen. VPCP regimen appears to be effective in the treatment of sqamous cell carcinoma of the head and neck.
