Application of Yeast DNA Microarray to Evaluate the Environmental Risk of Leachate and the Treated Leachate of Waste Disposal Site

Genomic responses to untreated and treated leachate from a waste disposal site were investigated using yeast DNA microarray. Differences were observed in both the number of genes induced and Munich Information Center for Protein Sequence (MIPS) functional categories after exposure to each of the landfill Leachate sample. Several of the genes related to homeostasis of metal ions were highly induced by exposure to leachate from the waste disposal site, while genes of unknown function were similarly induced by exposure to the treated leachate (treated using bacterial bed method and sedimentation with coagulation) . This results could be used to infer the nature of the substances in each of the landfill leachate sample. This study further demonstrated that DNA microarray analysis is a powerful technique for elucidating the toxicity of complex mixtures such as landfill leachate. We also demonstrated the potential application of DNA microarray analysis to the evaluation of the effectiveness and efficiency of certain waste treatment systems by characterizing gene expression profiles.
