Effects of Platelet-Derived Growth Factor(PDGF) and Basic Fibroblast Growth Factor(bFGF) on Wound Healing of Bone Defects.

The purpose of the present study was to evaluate the early wound healing events of bone defects which were made on the femora of beagle dogs with or without the application of recombinant platelet derived growth factor BB (PDGF) or basic fibroblast growth factor (bFGF). The dogs were killed 2 days, 1, 3 and 7 weeks following the application of 100ng of PDGF or bFGF in a methylcellulose gel. Control sites received the gel alone. Histopathological analyses of control specimens revealed no sign of wound healing around bone cavities 2 days post-operation, and no osteoid formation in spite of the presence of numerous osteoblast-like cells after 1 week. In contrast, growth factor treatment sites exhibited vasodilation and capillary proliferation 2 days post-operation, and fibrous granulation tissues were present and osteoid formation was observed after 1 week. In the control sites, new fine trabecular bone was observed on the wall and bottom of the cavity. After 3 weeks the cavity was found half filled, and the whole cavity after 7 weeks. In the growth factor sites, new trabecular bone was seen in the whole cavity after 3 weeks, and thick trabecular bone and lamellar bone were observed after 7 weeks. Tetrachrome osteoid staining exhibited a greater amount of calcified trabecular bone within the bFGF treatment sites compared to the PDGF sites after 3 and 7 weeks. These findings suggest that application of PDGF or bFGF can stimulate bone regeneration, and bFGF is much more effective than PDGF for bone wound healing.
