In the 1st report, the fibers of the depressor mandibulae muscle showed unimorphology of fast fibers by electron microscopy. Conversely, in the 2nd report, fibers were classified functionally into 3 groups by K<SUP>+</SUP>-sensitivity. It was indicated that the mechanism underlying the functional difference was in the triadic junction, also being the site of excitation-contraction (E-C) coupling. Thus, we speculated that functional difference in fast fibers was caused essentially by the functional difference in E-C coupling.<BR>The present study investigated the mechanism underlying the functional difference between fast fibers, using the same method as when investigating E-C coupling. Concanavalin A-ferritin (Con A-F) was used as a tracer to investigate the movement of electrometrin, a key protein involved in E-C coupling.<BR>It was revealed that (1) Con A-F binds a site on the T-tubular membrane opposite the foot, the site of E-C coupling; (2) depolarization evokes an outward movement of the Con A-F in the T-tubular lumen at this site; (3) the degree of contraction depends on that of movement; and (4) this site in the less K<SUP>+</SUP> sensitive group was located more centrally in the T-tubular lumen and was less sensitive to depolarization.<BR>These findings suggested that electrometrin also exists in the depressor mandibulae muscle as well as another skeletal muscle. It also suggested that the functional difference among fast fibers is caused by difference in the movement of electrometrin involved in E-C coupling.
