A new type of amphipathic antigen was isolated from A. viscosus ATCC 19246 by formamide extraction and partially characterized<BR>Amphipathic antigen was purified with Sepharose 6 B gel column chromatography. Gel diffusion test between the antiviscosus serum and amphipathic antigen prepicitated clearly in a single band.Chemical analysis indicated that hexose (31.4%), myo-inositol (6.8%), glycerol (7.1%), phosphorus (3.4%), protein (1.4%), fatty acid (31.5%) were components of amphipathic antigen. As the molar ratioof glycerol to fatty acids approximates 1: 1.8, this antigen may be diglyceride. Mild deacylation procedure, which removed the fatty acid residues (palmitic acid, stearic acid and oleic acid), destroyed the erythrocytes-sensitizing ability and immunogenecity together with depolymerization. Sugar on acid hydrolysis determined as alditol acetate derivatives by Glc and Glc-MS yielded four monosaccharides that correspond to mannose, galactose, glucose and myo-inositol (molar ratio, 1.0: 2.5: 1.0: 1.0). Other neutral sugar and amino sugar were not detected. Deacylated amphipathic antigen followed by dephosphorylation procedure converted into trisaccharide (mannose, glucose, myo-inositol, approximate molar ratio, 1: 1: 1) and monosaccharide (galactose, glycerol), inasmuch as phosphorus is conjugated across trisaccharide to monosaccharide. Repeating this structure forms a high molecular amphipathic antigen.
