The fine structure of the main excretory duct (MED) of parotid gland of the Japanese monkey, <I>Macaca fuscata</I>, was investigated by use of both scanning and transmission electron microscopies. It was observed that the MED epithelium consisted of light cells Types I, II and III, goblet and basal cells.<BR>Light cells Type I, II and III had no basal infoldings, but they had well-developed interdigitation between the neighboring cells. Numerous vesicles, which seemed to be pinocytotic, were observed in the cytoplam of light cell Type I immediately beneath the lumen. Abundant microtubules running parallel to the cell axis were seen in their infranuclear cytoplasm. There were also nemerous vesicles containing amorphous substance in the apical cytoplasm of light cell Type II. In their infranuclear cytoplasm, sacks of rough endoplasmic reticulum were abundant. Light cells Type III seemed to be degenerated cell.<BR>Goblet cells were full of mucous granules varied in electron density.<BR>Basal cell which were located in the basal fourth of the cell layer composing the MED epithelium, were small and nuclear ratio to cytoplasmic volume was high. They seemed to be stem-cells.<BR>The MED epithelium of parotid gland of the Japanese monkey did not seem to be involved in rendering the hypotonic saliva. However, the presence of goblt cells suggested that the MED epithelium seems to be involved in rendering the final saliva by adding mucous substance.<BR>Tuft cells and dark cells were not observed in the MED epithelium of the Japanese monkey.
