Water-insoluble extracellular materials synthesized in vitro by <I>Streptococcus mutans</I> of serotype a to g was examined on two different dried specimens by scanning electron microscopy.<BR>Air-dried specimens revealed that serotype a to e grown in sucrose-containing Trypticase Soy broth produced sticky amorphous capsule-like extracellular polysaccharide and serotype f and g produced massive extracellular materials. Serotype e to g also produced the extracellular materials which seemed to play a role of adherence to smooth surface in sucrose-free broth.<BR>In contrast to air-dried specimens, ethanol-dehydrated specimens visualized relatively smooth cell surface of serotype a to d grown in sucrose broth. Filamentous net-like structures by which many cells were assembled together were observed in the ethanol-dehydrated specimens of serotype b and d. Similar net-like structures were also observed on the cell surface of serotype g grown in sucrosefree broth, whereas massive materials as well as net-like structures were observed in sucrose-containing broth.<BR>These findings suggest that air-dried specimens may be suitable for morphological observation of intact insoluble extracellular polysaccharide of <I>S. mutans</I> and that significant differences in morphology of the extracellular materials observed among <I>S. mutans</I> serotypes may reflect differences in extracellular polysaccharides which they produced.
