Separation and characterization of two types of cell-associated glucan from Streptococcus mutans PK 1

Cell-associated glucan of S. mutans PK 1 was separated into two fractions by a mild mechanical treatment, gentle homogenization or weak sonification, and subsequent centrifugation. One of them (AG-1) was easily released from the cells by the treatment, but the other (AG-2) was not. AG-1 was a major constituent of the cell-associated glucan (about 90%) and AG-2 was a minor one. These two glucan fractions were characterized by periodate oxidation, methylation analysis, and Sepharose-CL-2B gel chromatography. AG-1 was predominantly composed of a high molecular weight glucan with a high proportion of α-(1→3)-glucosidic linkage. AG-2 mainly consists of a low molecular weight glucan with a high proportion of α-(1→6)-glucosidic linkage. AG-1 seems loosely associated with the cells, whereas AG-2 might be tightly associated with the cells.<BR>The cell-associated glucan of a strain FIL of S. mutans similarly separated into two types of glucan, whereas a mutant of S. mutans PK 1 defective in insoluble glucan synthesis and S. sanguis NCTC 10904 synthesized no loosely-associated polysaccharide.
