Effects of the Anti-allergic Agent (Mizolastine) in Guinea Pig Model of Allergic Rhinitis.

We studied the suppressive effect of an anti-allergic agent (Mizolastine) on nasal symptoms (sneezing and scratching movement), changes in toal airway resistance (TAR) and eosinophil infiltration induced by the topical antigen challenge in guinea pig models of allergic rhinitis. We also studied the suppressive effect of another anti-allergic agent, Terfenadine, on these responses. Mizolastine (1, 3mg/kg) and Terfenadine (10mg/kg) inhibited nasal symptoms induced by the antigen. We noted an dual phase increase in TAR induced by antigen. Mizolastine (3mg/kg) and Terfenadine (10mg/kg) partially but significantly inhibited the early phase increase in NAR. Mizolastine (3mg/kg) inhibited the later phase increase in TAR but Terfenadine (10mg/kg) did not affect this response. Mizolastine (1, 3mg/kg) inhibited eosinophil infiltration into the nasal mucosa induced by the antigen but Terfenadine (10mg/kg) did not affect them.In conclusion, Mizolastine inhibited nasal symptoms, the increase in TAR and eosinophil infiltration induced by the antigen in guinea pig models of allergic rhinitis. Therefore it is suggested that Mizolastine may affect nasal symptoms and, especially, nasal congestion in patients with allergic rhinitis.
