Ectopic Mineralization of Hen Leg Tendon.

Leg tendons of hens older than one year show ectopic mineralization recognized microscopically, and this symptom is already apparent in a 100-day old hen by electron microscopy. In the present study, we examined the fine structures of the minerals and organic matrices in the mineralizing front of hen leg tendon. The specimens were coated with osmium by glow-discharge plasma-polymerized film method and were observed by scanning electron microscopy. As a result, we found two novel structures at the mineralizing front area. First, there were elliptical mineralizing substances measuring 1.86±0.38μm (mean±S.D.) in length and 1.05±0.16μm (mean±S.D.) in width. These substances were fused to each other, forming straight rod-like structures. Moreover, transmission electron microscopy revealed that the surface of the elliptical structures had traces of a periodic pattern of 67nm, indicating that the mineral particles were surrounded with collagen fibrils. Secondly, we observed a wavy structure of collagen fibrils with regular periodicity of 2.8±0.38μm. Interestingly, a transition phase with straight fibrils converted to wavy structures was occasionally observed, and in such cases, the wavy and straight fibril structures were located on the mineralizing and unmineralizing sides, respectively. These findings strongly suggest that the formatio of wavy structures by collagen fibrils triggers the generation of mineral particles in the hen leg tendon during the ectopic mineralization process.
