Experimental study on the effects of calcitonin on mandibular invasion by VX2 carcinoma transplanted into gingiva in domestic rabbits.

It is important to determine the extent of invasion to the mandible of carcinoma cells in the treatment of gingival cancer. VX2 carcinoma cells were transplanted into the gingiva in domestic rabbits, and eel calcitonin (ECT) was injected into the thigh muscle. The invasion of VX2 carcinoma cells into the mandible and the effect of ECT were investigated by soft X-ray radiography, light microscopy, and scanning electron microscopy (SEM). In the early phase of bone invasion by VX2 carcinoma, the proliferation of osteoclasts played an important role in the resorption of the bone surface. However, 4 weeks after VX2 carcinoma transplantation, bone resorption was related to the proliferation of the VX2 carcinoma cells, and the number of osteoclasts decreased. SEM observation revealed the difference between the surface of the lacunae resorbed by osteoclasts and the surface of the bone resorption by VX2 carcinoma cells. Soft X-ray radiographs showed that ECT injection inhibited bone resorption by VX2 carcinoma cells. SEM revealed that the ruffled borders of osteoclasts consisted of long finger-like processes and plate-like processes. After ECT injection, the ruffled borders and the surface of the resorption lacunae underwent considerable changes. The ruffled borders of osteoclasts treated with ECT consisted of short plate-like processes.
