Study on the biocompatibility of hydroxyapatite by an osteogenic MC3T3-E1 cell line.

In order to examine the effects of hydroxyapatite on adhesiveness, growth and differentiation of bone-forming cells, clone MC3T3-E1 cells, which have retained osteoblastic characteristics, were cultured on smooth and rough-surfaced hydroxyapatite ceramics (HAC), and on smooth and roughsurfaced polysulfone resin (PSF) and plastic cover slips (PCS) as controls. On the smooth-surfaced HAC and PSF, the cells adhered well and showed flat morphology, and similar results were obtained on PCS. In confluent state, the cells showed polygonal-shaped morphology. In contrast, the cells always showed spindle-shaped morphology on the rough-surfaced HAC and PSF. On the smoothsurfaced HAC, the maximum cell number was obtained in all cultures at 3 days. At 15 days, alkaline phcsphatase activity, DNA and protein contents in the cultures on both the smooth and rough-surfaced HAC were significantly greater than thcse in cultures on both the smooth and rough-surfaced PSF. These results indicate that HAC accelerates the growth and differentiation of MC3T3-E1 cells. The cellular adhesiveness to HAC and PSF is affected by their surface structures.
