An ultrastructural study of the distribution of microtubules in osteoclasts.

The distribution of microtubules in usual active-osteoclasts, more active-osteoclasts treated with parathyroid hormone (PTH) and non active-osteoclasts treated with calcitonin (CT) incident to ex-perimental tooth movement in adult Wistar rats were studied.<BR>PTH and CT were injected intraperitoneally to each group of rats, 5 UPS/gbw at 10-30 minutes and 200 mU/gbw at 15-30 minutes before fixation, respectively. All of the animals were fixed by perfusion with 2 %glutaraldehyde containing 0.25 % tannic acid. 1). In the ruffled-border areas of usual and PTH-treated osteoclasts, microtubules were not quite observable. In the translated areas from clear zone to ruffled-border, bundles of microfilaments perpendicular to bone surface and bundles of other microfilaments oriented at right angle to their microfilaments were observed markedly.<BR>On the other hand, in the CT-treated osteoclasts, ruffled-border was not seen in the cytoplasm on the bone side, but microtubules and aggregates of microfilaments running parallel to the cell membrane in their cytoplasm were recognized clearly. This seems to be a "plasmalemmal undercoat". 2). In the central areas, network structures were formed by many microtubules in the cytoplasm of all osteoclasts. 3). In the vascular side of all osteoclasts, microtubules and microfilaments were running parallel to the cell membrane, and "plasmalemmal undercoat" was formed. These findings suggest that microtubules in osteoclasts may play an important role as their cytoskeleton.
