The therapeutic efficacy of TP-101 cream containing 1% miconazole nitrate (MCZ), which was designed for once daily application, was evaluated clinically and mycologically in a Trichophyton mentagrophytes-infected guinea pig model. The MCZ levels delivered to the stratum corneum of rat abdominal skin following topical application were measured using HPLC and the efficacy and delivery compared with those of commercially available 1% MCZ cream. The amounts of MCZ delivered by TP-101 were obviously larger than those from 1% MCZ cream, and 24hr after application were 2.4 times greator (p<0.01). The therapeutic efficacy of TP-101 was superior to that of 1% MCZ cream (p<0.10) both clinically and mycologically in the infected guinea pig model. These findings suggest that modification of the vehicle of antimycotic agents can increase the amount of drug delivered to the stratum corneum and provide good therapeutic efficacy in the guinea pig model of dermatophytosis.
