Combination Effect of Salicylic Acid on Antifungal Activity of Tolnaftate Preparation (ZM-589).

The antifungal agent, ZM-589 is a topical solution containing 2% Tolnaftate (TN) and 10% Salicylic acid (SA) as active ingredients.<BR>The minimum inhibitory concentrations (MICs) for TN, SA and their combination were determined here using ten strains among five species of dermatophytes. The therapeutic effect of ZM-589on experimental cutaneous<I>Trichophyton mentagrophytes</I>infection in guinea pigs was compared with ZM-SA (ZM-589 without SA), ZM-TN (ZM-589 without TN) and three commercial products: 1% Miconazol nitrate solution, 1%Ciclopirox olamine solution and 5% Exalamide solution (containing 2% Salicylic acid).<BR>MICs of TN and SA were 0.06-0.2 and around 1250μg/m<I>l</I>, respectively. MIC of the combination of TN and SA was essentially the same as that of TN alone.<BR>In a comparative study on experimental dermatophytosis on guinea pig skin, ZM-589 was statistically superior to any preparation used in the study including ZM-SA. The results were also supported by data on the negative rate of culture.<BR>These results suggest that ZM-589 can be beneficial in the clinical treatment of patients with dermatophytosis.
