Recent topics concerning the host-parasite relationship in some fungal infections were discussed. The main topics were as follows: (1) How are dermatomycoses (ex. Candida albicans, various species of Trichophyton and Microsporum) able to invade the stratum corneum?, (2) Why do dermatophytes rarely infect the scrotum?, and (3) Why dose depigmentation (or hypopigmentation) often occur in the lesions of Pityriasis versicolor? We tried to solve these problems through biochemical experimentation. The results obtained were as follows. (1) In media containing human stratum corneum as the nitrogen source, certain strains of Candida albicans produced keratinolytic proteinase. The purified enzyme had a pH optimum at 4.0 and belonged to the gruop of carboxyl proteinases. Biochemical properties of this keratinolytic proteinase were apparently different from those of the proteinases produced by some species of Trichophyton and microsporum. These results suggest that various kinds of dermatomycoses invade the stratum corneum by producing species specific keratinolytic proteinases. (2) It is thought that the reason for this is the high content of free fatty acid contained in the skin surface lipids. Comparative quantitative analyses of skin surface lipids were done on the thigh, back and scrotum regions. The content of free fatty acid from the scrotum was found to be of a much lower level than that from the other regions. Among the various kinds of dicarboxylic acids, C9-14 derivatives had an inhibitory effect on tyrosinase activity. These derivatives of dicarboxylic acid, which may possibly be related to the mechanism of depigmentation, were detected very frequently in the scales of the affected lesions.
