Comprehensive information on the morphological influence of various culture conditions in the presence of antibiotics and antimycotics to the formation of mitochondria in yeast cells is briefly described. Some aspects of ultrastructural information on the action mechanism of imidazole antimycotic, econazole, and aculeacin A are also introduced. The ultrastructure bases on the fungicidal action of membrane active antifungal agent econazole in Saccharomyces cerevisiae were explored by a combination of freeze-fracture and ultrathin sectioning electron microscopy. Econazole caused structural alterations in both fractured faces, particularly in the PF face, of the cell membrane, and it was characterized by a decrease in compactness of intramembranous particles. The results of the present study appear to meet the criteria based on biochemical studies that fungicidal level of econazole severely damages fungal membranes throuth the interaction with membrane phopholipids. In growing cells of Candida albicans, aculeacin A markedly inhibited the formation of normal cell walls. C. albicans protoplasts were regenerated in an osmotically stabilized liquid medium with or without aculeacin A. Untreated control protoplasts synthesized microfibrills on their surface. They were aggregated to thicker bunds which were intermeshed, forming a wide-meshed network of long fibrils. This microfibrillar structures probably represent chain of β-1, 3-glucan because they were broken down after treated ofβ-1, 3-glucanase and morphologically idendical microfibrils were synthesized in vitro by a cell-ree system in which fraction was used as a source of β-1, 3-glucan synthetase and UDP-glucose as a sole substrate. By contrast, in the presence of aculeacin A, the formation of normal fibrillar nets or bunds was significantly inhibited with resultant occurence of short fibrils. These electron microscopical data suggest that aculeacin A inhibits not only the synthesis of β-1, 3-glucan but the aggregation of microfibrils of this polysaccharide to form the crystalline structures.
