Biochemical studies on the mechanism of action of several minor antifungal chemotherapeutics, such as variotin, haloprogin, naftifin and tolnaftate, that preferetially inhibit filamentous fungi with only slight activity against yeast like fungi were made using the systems of: (i) yeast-phase cells of S. schenckii, (ii) arthroconidia of T. rubrum; and (iii) S. cerevisiae cell-free systems for macromolecular syntheses, especially DNA polymerases, RNA polymerases and cell wall polysaccharide synthetases. The results show that these drugs specifically and selectively inhibit synthesis of nucleic acids (DNA or RNA) and/or cell wall polysaccharides (β-glucan or chitin). It is also demonstrated that aculeacin A selectively inhibited the yeast β-glucan synthetase reaction, but did not the yeast chitin or mannan synthetase reactions.
