The relationship of 5-FC disc sensitivity to MIC on Mycoplate or on yeast morphology agar (YMA) plate was studied with several pathogenic fungi. With Cryptococcus neoformans, Aspergillus, and Candida, the first order correlation between inhibition diameter by 1μg 5-FC disc and log MIC was obtained (correlation coefcient=0.934 for Candida spp., 0.701 for Cryptococcus neoformans on Mycoplate, 0.541 for Cryptococcus neoformans on YMA, 0.543 for Aspergillus spp. on Mycoplate, and 0.453 for Aspergillus spp. on YMA). This correlation enabled a simple MIC determination from the inhibition diameter by 5-FC disc with these strains. Inhibition diameter of 1μg 5-FC disc against Cryptococcus neoformans and Aspergillus spp. on YMA was slightly larger than that on Mycoplate, while in the case of Candida spp., the inhibition diameter of 5-FC disc on YMA was similar to that on Mycoplate. On the other hand, in the case of Phialophora spp., MIC could only be roughly estimated by the comparison of the extent of growth among control, 1μg 5-FC disc and 100μg 5-FC disc containing agar plates.
