The inhibition of mitochondrial respiration by flavoglaucin from Aspergillus chevalieri

The inhibition mechanism of flavoglaucin, a hydroquinone mycoioxin from Aspergillus chevalieri on mitochondrial respiration was investigated using electron transport particles (ETP) which are reverted membrane vesicles of mitochondrial inner membranes. Flavo-glaucin was found to strongly inhibit succinate oxidase in ETP, indicating a direct interaction of flavoglaucin with enzyme(s) in the respiratory chain. Of the enzyme complexes in the respiratory chain, succinate dehydrogenase (complex II) and cytochrome c oxidase (complex IV) were not affected but succinate cytochrome c reductase (complex II, III) was strongly inhibited by flavoglaucin. These evidences clearly demonstrate that flavoglaucin specifically interfere with the redox reaction of complex III (cytochrome bc<SUB>1</SUB> segment) in the respiratory chain, which interpretes the mechanism for the inhibition of flavoglaucin on state 3 respiration in intact mitochondria.
