Change of Sensitivity of Bacteria against Ultra-violet Ray Radiated Repeatedly and Their Photoreactivation.

Three stains of Escherichia coli, 2 of Pseudomonas stutzeri, each 1 of P. aeruginosa, Staphylococcus epideremidis, S. aureus and S. hyicus recovered from working boots were used in Experiment 1. A volume of 0.02ml of hart infusion (HI) broth cultured these bacteria were dropped on test disk 5-8mm in diameter and dried in a desiccator. The disk was contaminated with from 104.5 to 106.3 of bacteria. The disks were exposed ultra-violet ray in a black box. Wave length of the ray was 250-260nm and amount of radiation of the ray was 104.75J/m2. The disks were soaked into tubes containing HI broth individually after the radiation whether bacteria on the disk contaminated survived or not. The HI broths in survived bacteria were collected and mixed in a flask. The test disks were made with the mixed broth survived bacteria. Radiation of the ray and making the disk were repeated 30 times.Escherichia coli, Pseudomonas stutzeri and P. aeruginosa were not recovered after 1st radiation of the ray and S. epideremidis also not recovered after 5th radiation. However, S. aureus and S. hyicus were still survived after radiation 30 times. The number of the test disks survived bacteria after each radiation fluctuated. No tendency was observed in bacteria to decrease sensitivity against ultra-violet ray.In Experiment 2, working boots after the radiation of the ray in black box were exposed natural day light during 24 hours to examine photoreactivation of the bacteria radiated ultra-violet ray. The number of bacteria contaminated in bottom of the boots just after radiation of ultra-violet ray and at 24 hours after the radiation were measured. A less number of bacteria detected at 24 hours after the radiation than that just after the radiation. No photoreactivation of bacteria was observed in bacterial counts.
