Effect of High Concentration of a Disinfectant Solution on Reduction of Viable Bacteria.

Experiments were conducted to evaluate the efficacy of concentration of 3 kinds of disinfectant solution. The disinfectant used in the experiments consisted of 10g/100ml of didecyl dimethyl anmonium chloride (invert soap), 1.75% of iodine (iodophor) and 70% of orthodichlorobenzene (ortho-drug), respectively. Dilutions of the disinfectants as routine use were 1:500 and 1:1, 000 of invert soap, 1:100 and 1:500 of iodophor, and 1:100 and 1:150 of ortho-drug. On the contrary, higher concentration of each disinfectant solution was 1:0 (non diluted), 1:1, 1:25 and 1:50. The plywood contaminated with bacteria in a chicken house were prepared for test pieces. A number of viable bacteria detected from the test pieces after washing were from 105.6 to 106.8/cm2. The test pieces were placed in air tight box of 1 m3 of capacity for spraying 500ml/m3 of the diluted disinfectant solutions.A number of 105.0-105.9/cm2 of bacteria were detected from the test pieces at 24 hours after spraying the disinfectant solutions diluted as routine use mentioned above. No statistical differences in bacterial counts detected from test pieces was observed among dilution ratio of each disinfectant and also no differences were observed among the 3 kinds of disinfectant. Reduction ratio of bacterial counts (bacterial counts before spraying/bacterial counts after spraying) was less than 10-2. Bacteria were not detectable when the same amounts of the undiluted disinfectant solutions were sprayed. After spraying invert soap and iodophol solutions diluted 1:25, bacteria reduced to less than 101/cm2, respectively. Bacteria reduced to less than 102/cm2 after spraying ortho-drug diluted 1:25. Reduction ratios of bacteria were 10-5.8, 10-6.1 and 10-4.8 by spraying invert soap, iodophol and ortho-drug, respectively. Results of the experiments suggest that the higher concentration of the disinfectant solution is required to reduce the more number of bacteria.Dilution ratio (concentration) of the disinfectant solutions for routine use has been depended on efficacy evaluated by phenol coefficient and minimum inhibitory concentration tested with bacteria in broth culture. It is considered that evaluation of efficacy of the disinfectant should be tested using bacteria in broth culture and also test pieces contaminated with bacteria under the natural condition.
