Determination of Corticosterone in Chicken Plasma by a Modified Method Using High-Performance Liquid Chromatography.

A modified method for the analysis of chicken plasma Corticosterone by highperformance liquid chromatography (HPLC) is described. One ml of plasma was mixed with 3ng of dexamethasone (internal standard) and 0.6ml of 0.25M sodium hydroxide. Then, the steroids were extracted with 15ml of dichloromethane. Dichloromethane fraction was evaporated and the subsequent residue was dissolved in mobile phase of the HPLC. Dexamethasone and corticosterone were determined by HPLC with UV detection at 248nm wavelength in retention times of 17 and 18min, respectively. Dexamethasone was useful not only as a tracer but also as a substance confirming the separation. Plasma corticosterone concentration of male broiler chickens (n=6) was estimated to be 2.15±1.00ng/ml (mean ± SD) by this method.
